How do you disconnect the transmission cooler line from the cooler ?

They're a little tricky, especially if they're corroded. I would spray them first and let them sit. The procedure is to hold the hose forward towards the cooler while pulling back on the plastic outer sleeve. Then pull back on the whole thing. Confusing at first, but you'll get it.
There are other very good write ups on how to do this. Make sure you wiggle the lines and plastic cover forwards and backwards a lot before you try this.

It can be a really big pain. I came close to cutting mine off but after many, many tries I was able to get it. Read up on the method and make sure you are doing right. Then keep working at it. I'll bet it took me a good hour of working with it to get mine off. Once off make sure you get the fitting back on all the way. Good luck
